I suppose for the sort of fishing I do (trout, running water and stillwater), if I had to restrict to one fly then it would be either a shaggy black klink/emerger type thing, or a small balloon caddis.

I would pick the following
Trout loch.....claret half hog
Silver.....cascade
River trout....cdc emerger
Sea trout.....big black and silver.

Lots of others needed for river trout in particular but hey Ho.

Running water only because I have no idea about the other stuff .Hans van Klinken - Lead head nymph , its a cased caddis thing . I use black tungsten beads instead of lead split shot.
By varying the weight ,it can be upstream dead drift or moved slightly .
Then fished down and across ect - used as a streamer .
Awful to cast and fish with but I would imagine catch fish any where .

a lot of cheating going on here.... one fly: not 4 or 5  :lol:  i would happily use a g & h sedge for carp & pike. albeit a bloody huge one for the latter  :8)
